Chimerism in Humans after Bone Marrow Transplantation: A New Challenge for Forensic DNA Experts
نویسنده
چکیده
When DNA profiling technique is being performed using any biological specimen from the individual, it promises the correct and unambiguous individualization, but in certain cases the forensic scientist may encounter the problem of mixed or completely mismatched DNA profile of a single individual [1,2]. STR’s (short tandem repeat) may lead to erroneous conclusion when the source of the biological material being analyzed is from a person who is a genetic chimera. In forensic genetics context, such genetic peculiarity may prevent the association of the perpetrator of an offence with the stain left at the crime scene or lead to false Identification of any suspect [2].

Clinical application of forensic DNA analysis: a literature review.
Polymorphic short tandem repeat (STR) analysis after polymerase chain reaction amplification is presently the most important analytical method of forensic DNA laboratory. Beside the forensic-purpose application, STR analysis is also necessary for chimerism testing after bone marrow or allogeneic stem cell transplantation. To increase success rate of the treatment and avoid adverse effect of the...
متن کاملRelationship between Molecular Chimerism and Graft Versus Host Disease after Allogenic Hematopoietic Stem Cell Transplantation
Background: Bone marrow transplantation (BMT) is considered as a curative therapy for a broad range of diseases. However, complications such as relapse and graft versus host disease (GVHD) may be observed following BMT. Chimerism analysis serves as a reliable indicator of transplant outcome. Complete chimerism refers to the complete replacement of hematopoietic system by donor cells, while mixe...

Mixed blood chimerism in T cell-depleted bone marrow transplant recipients: evaluation using DNA polymorphisms.
We have used DNA sequence polymorphism analysis to document engraftment after T cell-depleted bone marrow transplantation (BMT), with a selected panel of four DNA probes. In contrast to nondepleted BMT recipients, the patients who received T cell-depleted marrow exhibited a mixed blood chimerism. This mosaicism was observed before graft failure or relapse in six patients. However, in five other...
متن کاملEvidence for engraftment of human bone marrow cells in non-lethally irradiated baboons.
BACKGROUND Prior to organ harvesting, an attempt was made to modulate the donor's immune responses against prospective xenogeneic recipients by infusion of "recipient-type" bone marrow. METHODS For this purpose, baboons conditioned with total lymphoid irradiation were given 6 x 10(8) unmodified human bone marrow cells/kg body weight with no subsequent treatment. RESULTS Animals survived unt...
